Volumetric fluorescence imaging combined with modifled optical clearing for Alzheimer’s disease pathology

Abstract

We present the modified UbasM optical clearing and its combination -with 3D microscopic and mesoscopic optical imaging techniques for AD pathology research and the corresponding drug development. After efficient 3D immunostaining and optical clarification for mm-thick brain slices, confocal laser scanning microscopy and optical projection tomography are used for volumetric imaging of AD-YFP and thy1-YFP mice at multiple spatial scales, showing a significant increase of amyloid—β plaques and the decrease of neurons in AD model.
