Abstract
Using a nanoaperture optical tweezer with two single frequency trapping lasers, we obtain the Raman spectra of proteins and note their interactions with small molecules after binding. This approach to acoustic Raman has high-sensitivity (single molecule) and high-resolution, while not suffering from the background of the zero loss line. We compare protein-ligand binding induced spectral changes to normal mode calculations.
