3D localization of fluorescent microparticles using a rotating point spread function

Abstract

We demonstrate single-image three-dimensional position localization of fluorescent microspheres embedded in a 3D volume by engineering the point spread function of a wide-field fluorescent microscope to rotate continuously with defocus.
