Dual-color super-resolution imaging by fluorescence emission difference microscopy

Abstract

Fluorescence emission difference microscopy(FED) has been demonstrated to be capable of breaking the diffraction limit through the difference of two different confocal images. Here we propose and build a novel dual-color FED system that can provide fluorescence imaging with sub-diffraction resolution. The enhancement in the lateral resolution is verified theoretically and experimentally. Compared to standard STED system, FED enjoys the advantages of lower illumination power and higher flexibility. In addition, the dual-color configuration enables the presented system to be suitable for biological investigations.
