Imaging actin filament network using random localized near-fields

Abstract

To overcome axial diffraction limit, total internal reflection fluorescence (TIRF) microscopy has been used for investigating biological studies including subcellular interactions between various biomolecules or organelles. However, its lateral resolution remains limited by diffraction limit. To address this issue, here, we have employed plasmonic silver nanoislands to create localized near-field and to use it to excite fluorescent molecules. Image deconvolution for super-resolution imaging based on pre-determined near-field distribution is also discussed [1,2].
