Effect of Imaging in Tissue on Resolution of Pulsed STED Two-Photon Excitation Microscopy

Abstract

We examine the resolution of two-photon imaging combined with STED microscopy and find resolution depends on imaging depth. The depth dependence of resolution is consistent with the depth dependence of depletion efficiency, suggesting resolution is limited by STED laser propagation through tissue.
