Optimized visual AuNP beacon for tracing mRNA changes in living cells

Abstract

In this manuscript, we prepared a highly efficent molecular beacon by manipulating the number of sequences on AuNPs and surface modification with PEG and cell-penetrating peptide. Meanwhile, the cellular uptake mechanism of the AuNP beacon was studied. Subsequently, we designed a whole set of processes to enhance the cellular uptake of our molecular beacon based on the mechanism, including up-regulation of scavenger receptors of cells and ultrasound treatment. Thus, we have established an integrated method to increase the detection efficiency and accuracy of a molecular beacon from synthesis modification and exogenous mediation to gene regulation.
