Measurement of Fluorescent Spectra of Liquids with a Modified Beckman DU Spectrophotometer

Abstract

A description is given of an attachment for the Beckman DU spectrophotometer for measuring the fluorescent spectra emitted by liquids and solutions. The external optical system normally used for absorption spectra measurements was modified to provide optimum fluorescent intensity with minimum interference from scattered and reflected primary radiation and from fluorescence of the cuvettes. Modifications were also made in the input circuit to permit use of a photomultiplier detector for increased sensitivity. Otherwise, the amplifying and dispersing systems of the instrument were used in the usual manner. Typical spectral distribution and analytical curves and sensitivity data obtained with this attachment are included.
