Abstract

The three-dimensional optical transfer function of a confocal fluorescence microscope is derived. It has no missing cone and provides tomographic images of the sample. The derivation is based on the dispersion equation of spherical waves and the diffraction limitation by an objective lens. Experimental results are shown to verify the derivation.
