Using the method of depth reconstruction from focusing for microscope images

Abstract

This paper discusses a method of constructing a depth map of a scene from a set of microscope images of the scene. For each coordinate of the image field, the number of the layer in which the variance over a window of definite size takes its maximum value is determined. It is proposed to preprocess the images before the stage of computing the variance, and the operation of separating the image of the object from the background is to be carried out by means of an adaptive threshold with respect to the difference of the maximum and minimum variance over all the layers for each pixel separately. Experimental studies show that the proposed method is suitable for constructing depth maps of various microscopic objects.
