Label-free protein detection using terahertz time-domain spectroscopy

Abstract

Quantitative analysis of protein is the basics to understand the mechanisms of biological processes in terms of the interactions, activities and regulation of the underlying proteins. Protein immunoblot belongs to the common methods to determinate protein content and includes several complicated and time-consuming steps. In the proposed method the membrane made of nitrocellulose after being moved on proteins is directly scanned by using terahertz time domain spectroscopy system (THz-TDS) to qualitatively and quantitatively protein species and quantity. It does not require the detected proteins being labeled by corresponding antibodies, which is free from tedious interlocking steps and much time is saved. Six different kinds of proteins under investigation can be distinguished by refractive index extracted from THz transmission spectrum.
