Determination of S-Collidine

Abstract

<i>S</i>-Collidine (2,4,6 trimethylpyridine) is used in some buffers for fixing the tissue for light and electron microscopy. Sometimes it is contaminated with mono and disubstituted products. It is therefore suggested to purify prior to use. The method for purification is difficult and it is not always necessary. This paper describes a simple method for assaying purity of <i>s</i>-collidine by reacting it with <i>p</i>-dimethylaminobenzoic acid (<i>p</i>-DMAB) in acidic alcoholic medium which gives a violet color immediately.
